I think I will uninstall it and start all over again. I won't lose much as I haven't done a lot with this yet.

I uninstalled Jamstix and verified folders were gone. Reinstalled 3.5.0 and still have the same issue. By the way, I'm only using the 64bit Jamstix plugin within Reaper 64bit.

Here are the steps I take to create the error in more detail:
Start Reaper 4.57/x64
Open Insert menu and choose "Virtual instrument on new track"
Choose JAMSTIX 3
Answer No to create a single stereo track
In Jamstix...
Click Kit, Load Rock Standard kit
Click Song
By default a verse part is already there with 4 bars and 30 reps
I change the default verse from 30 reps to 1 rep
I add a single RH on beat 1 of each bar (just to place something in the groove)
Open the bar edit menu
Choose Save Groove As Style
Give it a name
Indicate 4 bars for groove, click OK
Runtime error 216 at 000000000975E045
Can you duplicate?

I get the same error in Sonar X2 Producer 64 bit.

Make sure the permissions on c:\jamstix3 are set to 'Everyone' and try again.

Set permissions to c:\jamstix3 Everyone Full Control. Same error. I verified the permissions were applied to everything below.

Does 'Save Bar' in the bar editor menu work or throw the same error?

Yes, "Save Bar" throws the same error.

I created a 30MB video showing how I produce the error. It is a swf file.
Would this be something that could help in figuring this out? If so, what is the best way for me to get this video to you? I have Google Drive and Dropbox accounts.
